強く長く続く免疫をつくるために
Toward Strong and Long-Lasting Immune Protection


私たちの目標は、強く長期間持続する免疫を生み出す基本原理を理解し、免疫応答を探究・制御するための新しい知識やツールを創出することです。

強い免疫はどのように生み出されるのでしょうか?

ワクチンによる免疫記憶はなぜ長期間維持されるのでしょうか?

なぜ免疫は病原体だけでなく、がん細胞も認識して攻撃できるのでしょうか?

そして、免疫細胞はどのように情報を読み取り、応答を制御しているのでしょうか?

私たちの研究室では、 「免疫はどのように情報を処理し、強く持続的な免疫応答を生み出すのか」 という基礎的な問いに取り組んでいます。

また、免疫システムを構成する分子を人工的に組み合わせることで、 「どのようなシグナルの組み合わせが免疫応答を制御するのか」 を理解しようとしています。

こうした研究を通して、将来の免疫療法やワクチン開発につながる免疫学の基盤を築くことを目指しています。(図1)


Our goal is to understand the fundamental principles that generate strong and long-lasting immunity and to develop innovative tools for exploring immune regulation.

How are strong and durable immune responses generated?

Why does vaccine-induced immune memory persist for years?

How can the immune system recognize and eliminate not only pathogens but also cancer cells?

And how do immune cells process information and regulate their responses?

Our laboratory seeks to answer these fundamental questions by studying how immune responses are generated, regulated, and maintained.

We also use synthetic biology-based approaches to investigate how combinations of immune signals influence immune cell function

Through these studies, we aim to generate new knowledge and tools for exploring immune regulation and to build a scientific foundation for future advances in immunotherapy, immune modulation, and vaccine development.

① 強い免疫はどのように成立するのか?

私たちが目指す「強い免疫」とは、

  • •がんや病原体を効率よく排除できる
  • •必要な時に適切に働く
  • •長期間記憶される

という特徴をもつ免疫です。(図2)。

このような免疫がどのように成立するのかを理解するために、私たちは

  • •T 細胞が活性化される仕組み
  • •活性化シグナルが細胞内で伝達される仕組み
  • •それらを制御する分子の働き

を分子・細胞・個体レベルで研究しています。

さらに、その知見をもとに免疫応答を人為的に制御する方法についても探究しています。


① How Is Strong Immunity Established?

The type of immunity we study has three important characteristics:

  • •Efficient elimination of cancer cells and pathogens
  • •Appropriate activation only when needed
  • •Formation of long-lasting immunological memory

(Figure 2)


To understand how such immune responses are generated and maintained, we investigate:

  • •How T cells become activated
  • •How activation signals are transmitted within cells
  • •How these signaling pathways are regulated

Using molecular, cellular, and animal-based approaches, we seek to uncover the fundamental mechanisms that govern T-cell-mediated immunity.

We also explore how this knowledge may contribute to future strategies for controlling immune responses.

② T 細胞とは?

体内を巡回しながら病原体や異常細胞を監視し、それらを認識すると免疫応答を開始します。

また、T 細胞は病原体やがん細胞を排除するだけでなく、一度遭遇した抗原を記憶し、同じ抗原に再び出会った際により迅速かつ強力に応答する能力を持っています。

この「免疫記憶」は、長期間持続する免疫防御を支える重要な仕組みであり、ワクチンの効果やがん免疫療法の基盤となっています。

私たちの研究室では、こうしたT細胞の活性化や免疫記憶形成の仕組みを理解することを目指しています(図3)。


② What Are T Cells?

T cells are key players in the immune system and are essential for generating effective immune responses.

They continuously patrol the body, monitoring for pathogens and abnormal cells. Upon recognizing a threat, they become activated and initiate immune responses to eliminate it.

In addition to protecting the body from infection and cancer, T cells can retain a memory of previously encountered antigens. This enables them to respond more rapidly and effectively when the same antigen is encountered again.

This ability, known as immunological memory, is a fundamental mechanism underlying long-term immune protection and forms the basis of vaccination and cancer immunotherapy (Figure 3).

Our laboratory seeks to understand how T-cell activation and immunological memory are established and maintained.

③ 二重シグナル理論とは?

T細胞は「敵を見つけた」だけでは動きません。

誤って自分自身を攻撃しないために、

  • •シグナル1:抗原を認識
  • •シグナル2:本当に攻撃すべきという許可

という2つの情報が必要です。

これを「二重シグナル理論」と呼びます(図4)。


③ What Is Two-Signal Theory?

T cells do not attack simply because they encounter an antigen.

To prevent accidental attacks against the body's own tissues, T cells require two distinct signals:

  • •Signal 1: Recognition of antigen
  • •Signal 2: Confirmation that an immune response should proceed

Both signals must be present for full T-cell activation.

This concept is known as the Two-Signal Theory of T-cell Activation (Figure 4).

④ 強い免疫を生み出すスイッチ「OX40」とは?

T細胞はどのようにして長期間働く免疫記憶を獲得するのでしょうか?

私たちは、その仕組みに関わる補助刺激受容体 OX40 に注目しています。

OX40 は活性化した T 細胞に発現し、T 細胞が受け取る「シグナル2」を増強することで、

  • •免疫応答の持続
  • •T 細胞の生存
  • •免疫記憶の形成

を促進します(図5)。

そのためOX40は、強く長期間持続する免疫を生み出す重要な分子と考えられています。


なぜ重要なのか?

OX40がどのようにT細胞の働きを調節しているのかを理解することは、

  • •がん免疫
  • •ワクチン応答
  • •免疫記憶形成

の基本原理を理解するうえで重要です。

私たちはOX40シグナルの仕組みを解明するとともに、OX40を標的とした人工タンパク質の開発を通じて、免疫応答を制御する新しい方法の可能性を探っています。

④ OX40: A Molecular Switch for Strong and Durable Immunity

How do T cells acquire long-lasting immunological memory?

To address this question, we focus on OX40, a costimulatory receptor expressed on activated T cells.

OX40 enhances Signal 2 received by T cells and promotes:

  • •Sustained immune responses
  • •T-cell survival
  • •Formation of immunological memory
(Figure 5)

Because of these functions, OX40 is considered an important regulator of strong and durable immune responses.


Why Is OX40 Important?

Understanding how OX40 regulates T-cell responses can provide important insights into the fundamental mechanisms underlying:

  • •Anti-tumor immunity
  • •Vaccine-induced immune responses
  • •Immunological memory

Our laboratory seeks to understand the molecular mechanisms of OX40 signaling and to explore how this knowledge may contribute to future approaches for controlling immune responses.

In addition, we develop and characterize engineered proteins targeting OX40 as tools for studying and modulating costimulatory signaling.

⑤ なぜ人工タンパク質が必要なのか?

免疫応答は、1つのシグナルだけでは決まりません。

T 細胞は複数の受容体からの情報を統合し、その結果として活性化や免疫記憶形成が起こります。

しかし天然の分子だけでは、そのような複数のシグナルの組み合わせを自由に調べたり制御したりすることは困難です。

そこで私たちは、複数の免疫シグナルを組み合わせた人工タンパク質を設計しています。


私たちの研究

私たちはT細胞を活性化する分子や増殖を促す分子を「部品」として捉えています。

例えば、

  • •OX40L
  • •4-1BBL
  • •GITRL
  • •CD70

といった補助刺激分子や、

  • •IL-2

のようなサイトカインを組み合わせることで、新しい機能を持つ人工タンパク質を作製しています。

これらの分子は天然には存在しない組み合わせを持ち、免疫細胞にこれまでにないシグナル環境を与えることができます(図6)。


なぜ重要なのか?

人工タンパク質は、免疫シグナルの組み合わせがT細胞の働きにどのような影響を与えるのかを調べるための新しい研究ツールにもなります。

私たちはこのアプローチを通して、

  • •T 細胞がどのように複数のシグナルを統合しているのか
  • •どのようなシグナルの組み合わせが強く持続的な免疫応答を生み出すのか

を理解したいと考えています。

こうした知見は、将来の免疫制御技術やワクチン開発につながる可能性があります。


⑤ Why Do We Need Artificial Proteins?

Immune responses are not controlled by a single signal.

Instead, T cells integrate multiple signals received through different receptors, and the combination of these signals determines how immune responses develop.

However, naturally occurring molecules provide only limited opportunities to investigate or control these signal combinations.

To address this challenge, we design artificial proteins that combine multiple immune signals into a single molecule.


Our Research

We view immune molecules as modular biological components that can be assembled into new functional designs.

For example, we combine costimulatory molecules such as:

  • •OX40L
  • •4-1BBL
  • •GITRL
  • •CD70

with cytokines such as:

  • •IL-2

to create artificial proteins with functions not found in nature.

These engineered proteins provide immune cells with novel signaling environments and serve as powerful tools for studying immune regulation (Figure 6).


Why Is This Important?

Artificial proteins allow us to investigate how combinations of immune signals influence T-cell behavior.

Using these molecules, we seek to understand:

  • •How T cells integrate multiple signals
  • •Which signal combinations promote stronger and more durable immune responses
  • •How immune memory is established and maintained

Our long-term goal is to uncover design principles for immune regulation.

Knowledge gained from these studies may contribute to future advances in immune modulation, vaccine development, and immunotherapy.

TRAF5による免疫シグナル制御機構の解明

⑥ なぜ TRAF5 は複数の受容体を制御できるのか?

TRAF5 は細胞内アダプター分子であり、複数の免疫受容体から伝わる情報を統合し、その強さや質を調節する「交通整理役」のような分子です。

私たちは、「なぜ1つの分子が多様な受容体の働きを制御できるのか?」という根本的な疑問に取り組んでいます。

免疫細胞には、

  • •CD40
  • •IL-6受容体
  • •IL-27受容体
  • •Toll様受容体

など多くの受容体があります。

興味深いことに、TRAF5 はこれら異なる受容体に共通して関与しています(図7)。

私たちの仮説

TRAF5の量そのものが、免疫応答の強さを決定している可能性があります。

もしこれが正しければ、TRAF5は単なる情報伝達分子ではなく、免疫応答全体の強さを決定する重要な調節因子と考えられます。

私たちの研究

私たちは TRAF5 に注目し、「受容体の違いを超えて免疫シグナルを調節する共通原理」を明らかにしようとしています。

TRAF5 は単なる情報伝達分子ではなく、免疫応答の強さや質を決める調節因子である可能性があります。

特に近年は

  • •CD40 シグナルによるB細胞活性化
  • •抗体産生や胚中心形成
  • •IL-27 シグナル

におけるTRAF5の役割に注目しています。


TRAF5の理解は、「強い免疫がどのように作られ、どのように制御されているのか」という根本的な問いの解明につながると考えています。

「TRAF5の量が免疫応答を決めるかもしれない」という研究室独自の問いに答えるために研究しています。

Mechanisms of Immune Signaling Regulated by TRAF5

⑥ Why Can TRAF5 Regulate Multiple Receptors?

TRAF5 is an intracellular adaptor protein that acts like a traffic controller that integrates and regulates signals from multiple immune receptors.

A fundamental question driving our research is:

How can a single molecule regulate signaling from many different immune receptors?


Immune cells express numerous receptors, including:

  • •CD40
  • •IL-6 Receptor
  • •IL-27 Receptor
  • •Toll-Like Receptors (TLRs)

Interestingly, TRAF5 participates in signaling downstream of all of these receptors (Figure 7).


Our Hypothesis

We hypothesize that the level of TRAF5 expression within immune cells may directly influence the strength of receptor-mediated signaling.

If this hypothesis is correct, TRAF5 may function not only as a signaling adaptor but also as a regulator of immune response magnitude.


Our Research

We focus on TRAF5 to uncover the common principles by which diverse immune receptors are regulated.

Rather than acting as a simple signaling adaptor, TRAF5 may serve as a key regulator that shapes both the strength and quality of immune responses.


Our recent studies have investigated the role of TRAF5 in:

  • •CD40-mediated B-cell activation
  • •Antibody production and germinal center formation
  • •IL-27 receptor signaling

Through these studies, we aim to understand how immune receptor signaling is coordinated across different signaling pathways.

In particular, we are investigating the possibility that the level of TRAF5 expression may influence the magnitude of immune responses.

Understanding how TRAF5 regulates immune signaling may reveal fundamental principles governing the generation and maintenance of strong and durable immunity.

私たちの研究室では、

「免疫はどのように情報を読み取り、応答を制御しているのか?」

また、免疫システムを構成する分子を人工的に組み合わせることで、

「どのようなシグナルの組み合わせが強く持続的な免疫を生み出すのか?」

を理解しようとしています。


私たちは、

How do combinations of immune signals generate strong and long-lasting immune responses?

By combining fundamental studies of immune signaling with synthetic biology-based approaches, we aim to:

  • •Uncover fundamental principles of immune regulation
  • •Develop new knowledge and research tools for exploring immune signaling

Through these efforts, we hope to build a scientific foundation for future advances in immunotherapy, immune modulation, and vaccine development (Figure 8).
